Job Details
Air-pollution is ranked as a Group 1 carcinogen by the International Agency for Research on Cancer. Nitroarenes are among the carcinogens responsible, however, the nitroreductases involved in their metabolic activation have not been fully identified. A postdoctoral position is available immediately to study the role of the nitroreductase activity of human aldo-keto reductases (AKRs) in the metabolic activation of nitroarenes. Studies involve measurement of DNA-adducts in bronchial epithelial cells with genetic and pharmacological manipulation of AKRs; induction of AKRs by the Nrf2-Keap1 pathway and determination of whether this exacerbates nitroarene activation; and examination of single-nucleotide polymorphisms in AKR genes and their impact on nitroreductase activity. The position is available in the laboratory of Trevor M. Penning in the Center of Excellence in Environmental Toxicology (CEET) at the University of Pennsylvania. A successful applicant can participate in a rigorous training program supported by training faculty in basic and clinical sciences.
Required qualifications: PhD within the last three years in the biomedical sciences (e.g. biochemistry, pharmacology, toxicology, or analytical chemistry).
Preferred qualifications: Strong background in enzymology, analytical chemistry using LC-MS/MS, mammalian cell culture, gene and protein expression (western blot analysis, real-time PCR, enzyme assays); use of si-RNA, sh-RNA and CRISPR/cas9 is desirable.
